A DNA result is only as reliable as the sample behind it. Whether you are seeking reassurance about a family relationship or preparing for a formal legal matter, knowing how to collect DNA samples correctly helps protect the accuracy, clarity and usefulness of your result.
For most at-home relationship tests, a simple cheek swab provides the DNA needed for analysis. The process is straightforward, but small mistakes - such as swapping envelopes, collecting too soon after eating, or touching the swab tip - can delay testing or require a new sample. When the outcome matters, careful collection offers valuable peace of mind.
Start by choosing the right type of DNA test
Before opening a kit, confirm whether you need a personal test or a legal DNA test. The sample collection method may look similar, but the purpose and process are very different.
A personal DNA test is generally used for private knowledge. Participants collect their own samples at home, following the kit instructions, and send them to the laboratory for testing. These results can provide clear answers about biological relationships, but they do not usually meet court, immigration or other official evidentiary requirements.
A legal DNA test requires identity verification, witnessed collection and documented chain of custody. This means an authorised collector confirms each participant's identity, takes the samples, completes the paperwork and ensures the sealed samples are transferred securely to the laboratory. If your results may be used for Family Court proceedings, inheritance matters, immigration, child support or another official purpose, arrange legal collection from the outset. A home-collected sample cannot normally be converted into a legal result later.
How to collect DNA samples correctly with cheek swabs
Your test kit will include specific instructions, and those instructions should always take priority. However, the following preparation and collection practices apply to most buccal, or cheek, swab DNA tests.
Prepare before you open the swabs
Collect samples in a clean, dry area with enough space to keep each participant's materials separate. If several people are being tested, work with one person at a time. This greatly reduces the chance of placing the wrong swab in the wrong envelope.
Ask each participant to avoid eating, drinking, smoking, chewing gum, brushing their teeth or using mouthwash for at least 30 minutes before collection, unless the kit states a different timeframe. Food residue, drinks and oral products can interfere with the quality of the sample. For babies, it is best to wait after a feed and gently wipe excess milk or food from around the mouth before swabbing.
Wash and dry your hands thoroughly. Then complete any labels or identifying details exactly as directed by the kit. Check the spelling of names and dates of birth carefully. A correctly collected swab can still cause delays if the paperwork is incomplete or participant details do not match.
Swab the inside of the cheek firmly
Open one swab without touching its collection tip. Hold it by the handle and rub the tip firmly against the inside of one cheek, rotating the swab as you go. Use enough pressure to collect cheek cells, not just saliva. Follow the number of strokes or collection time stated in your kit, as laboratories may use different swab types and procedures.
If the kit includes more than one swab per participant, use each one as instructed. Often, this means swabbing both cheeks or repeating the process with a second swab. Multiple swabs give the laboratory sufficient DNA and support reliable analysis.
Do not allow the swab tip to touch your hands, bench, clothing or any other surface. Do not share swabs between people, even within the same family. Each swab must be used for one named participant only.
Let the swabs dry and package them properly
Many collection kits require swabs to air-dry before they are placed in their labelled paper envelope or collection sleeve. This helps prevent moisture from encouraging mould or degrading the sample during postage. Never use a hair dryer, heater or direct sunlight to speed up drying, as excessive heat can damage DNA.
Only place swabs in the packaging supplied with the kit. Avoid plastic bags unless they are specifically provided and directed for use, because trapped moisture may affect the sample. Once packaged, seal each participant's envelope carefully and check the labels again before placing them in the return satchel.
Do not mix separate participants' swabs in one envelope. Keeping every sample clearly separated is one of the simplest ways to avoid administrative errors and protect the integrity of the testing process.
Prevent contamination and mix-ups
Human DNA is common in everyday environments. A stray touch, a reused surface or an unlabelled envelope can introduce avoidable uncertainty. The laboratory has quality controls, but careful collection gives testing the best possible starting point.
Use a fresh, clean surface and keep pets away from the collection area. Do not collect samples immediately after someone else has handled the participant's mouth, particularly for young children. If a parent has just applied lip balm, wiped a child's mouth or helped with food, wash hands and allow time before collecting the swab.
For infants and young children, an adult can perform the swab, but should handle only the stem. Calm collection is usually easier when the child is comfortable and has not just eaten. If a child will not tolerate the swab, do not force the process. Contact the testing provider for guidance on the most suitable next step.
If you accidentally touch the swab tip, drop it, use the incorrect label or are unsure which participant it belongs to, do not guess. Set it aside and request a replacement collection device or kit. Retesting is preferable to proceeding with a sample whose identity or quality is in doubt.
Follow timing and return instructions
Once samples are dry, return them as soon as practical using the method supplied with your kit. Do not leave completed samples in a hot car, a damp bathroom or direct sunlight. If you cannot post the kit immediately, store the sealed materials in a cool, dry place away from moisture and heat.
Include all required forms and ensure declarations are signed where requested. Missing consent forms, incomplete participant details or incorrect sample identifiers can pause processing even when the DNA itself is suitable for testing.
It is also wise to retain any lodgement receipt or tracking information after posting. This is especially helpful if you are working to a deadline or need reassurance that the samples have reached the laboratory.
When cheek swabs are not the right option
Cheek swabs are the preferred sample for most relationship testing because they are non-invasive, reliable and easy to collect. Other items, such as hair, toothbrushes, razors or personal effects, are not interchangeable with a standard cheek swab and should not be submitted without laboratory advice.
These non-standard samples can carry a higher risk of contamination, may contain little usable DNA and can require specialist testing methods. They are sometimes considered in DNA reconstruction or complex kinship matters where a person is unavailable, but suitability depends on the item, its history and the question being investigated. A laboratory should assess the circumstances before you collect or send anything.
Prenatal paternity testing also follows its own clinical process. It should be arranged through an appropriate provider rather than attempted through a standard home collection kit.
Consent, privacy and legal collection
DNA testing involves sensitive personal and family information. Every adult participant should understand the test being undertaken and provide the required consent. For a child, consent requirements may depend on parental responsibility, guardianship arrangements and the purpose of the test. If there is disagreement between parents or an active legal dispute, seek appropriate legal guidance before proceeding.
For legal testing, do not attempt to collect samples privately. The chain of custody is central to the result's legal standing. An independent collector will check photographic identification, take photographs where required, witness the collection and seal the samples under controlled procedures.
